If this Effect were on City Wall would they mutually protect each other, thus making their controller's postition absolutely unassailable, and resulting in a game winning lock down?

Almost, however, there are ways to deal with the City Wall without taking it down. You can also deal with the Effect without targeting it, like with a Cleanse for instance. Plus, any spell or effect (able to) can target something protected by a wall so long as the casters have flying. This opens up a whole bunch of ways to deal with this scenario. But it very well could be a winning combo if your opponent is not ready for it.
